Medulloblastoma is the most common malignant brain tumor diagnosed during childhood. Recent advances in radiation and chemotherapy improved the survival, but an important rate of the medulloblastoma patients have been died because of a rapidly progressive disease. Newly described research techniques in the field of molecular biology and genetics and the opportunities provided by these techniques to the researchers allowed advanced understanding of the mechanisms of every disease in every medical field. By the means of these techniques, researches on the pathogenesis of central nervous system (CNS) tumors carry on worldwide. These researches had emerged different approaches for every single tumor and keep on emerging. Medulloblastoma is one of the most studied CNS tumor in terms of molecular biology and genetics contemporary, and molecularly subdivided into four groups (WNT, SHH, Group 3, Group 4) which this classification has provided us having a prediction on the prognosis of the disease. Future treatment protocols will be associated with this molecular classification. Ongoing researches will aid further understanding of basic mechanisms of the disease and also important for new targeted therapies. In this paper, we briefly reviewed the advancements of medulloblastoma pathogenesis provided by recent molecular biology and genetic researches.
