A chordoma originates from remnants of the embryonic notochord and is located along the axial skeleton. This is a tumor of the skeleton that is usually low grade and is made of calcified lobules within the bone. This paper aims to review current molecular biological advances on the tumor-biology, diagnosis and treatment of chordomas. Advances in chordoma molecular biology are reviewed under different headings corresponding to various processes of cellular biology. Genetic and molecular biological findings of various study groups and the current take on these findings are summarized. Our understanding on the origin and development of chordomas has increases substantially after the application of molecular biological technology. However, these advancements have not yet resulted in novel and effective treatment strategies. Further studies are required to design effective treatment strategies for chordomas.
