Experimental studies in the last ten years demonstrated that excitatory amino acids play an important role in the pathogenesis of many central nervous system diseases. Experimental studies with excitatory aminoacid antagonists have been found to be beneficial in various models such as cerebral ischemia, head and spinal cord injury, epilepsy and some chronic degenerative diseases. In this article we reviewed our experience with excitatory amino acid neurotoxicity gained from experimental studies with various models (focal cerebral ischemia, head and spinal cord injury) carried out in our research laboratories. We also reviewed the current medical literature and aimed to give a brief summary on the neurotoxic mechanisms of important central nervous system diseases and therapeutic applications.
