Tadalafil on Vessel Structure and Apoptosis in Vasospasm After Experimental Subarachnoid Hemorrhage

DOI : 10.5137/1019-5157.TND.3298 AIM: The aim of this study is to investigate the effectiveness of tadalafil, a selective and reversible phosphodiesterase-5 (PDE-5) enzyme inhibitor, on the vasospasm developed after experimental subarachnoid hemorrhage (SAH).

MATERIAL and METHODS: Rats are divided into 5 groups as control saline, SAH, SAH + tadalafil 5 mg/kg, SAH + tadalafil 10 mg/kg and SAH + tadalafil 15 mg/kg. In control saline group; saline was given into subarachnoid space through cistern magna and tadalafil solvent was administered intraperitoneally. Autologous arterial tail blood was given into subarachnoid space through cisterna magna in SAH and SAH + all tadalafil groups. In SAH group, only tadalafil solvent was applied intraperitoneally whereas in SAH + tadalafil groups tadalafil was applied at different dosages (5 mg/kg, 10 mg/kg, 15 mg/kg) intraperitoneally. Rats were sorted as 3rd day and 7th day and decapitated on the stated day. Basilar artery wall thickness, lumen space and the quantity of apoptosis were examined in all groups.

RESULTS: Tadalafil at all dosages reduced vasospasm when examined by vessel wall thickness and lumen space at 3rd and 7th days. Tadalafil reduced early apoptosis (3rd day) at all dosages, meanwhile late apoptosis (7th day) was reduced only in high dose (15 mg/kg) group.

CONCLUSIONS: Tadalafil has shown to be beneficial in treatment of cerebral vasospasm following experimental SAH in our study. If further investigations support our results, tadalafil may be a useful agent in treatment of aneurysmal SAH.
